Whetton was born in Melbourne, Victoria, on 5 January 1958. She earned the degree of a Bachelor of Science (Honours) majoring in Physics as well as an honours year in Meteorology at Melbourne’s University of Melbourne. She earned an honorary Doctor of Philosophy degree from the same institution in the year 1986.

Whetton resided within Footscray, Victoria, with her spouse Janet Rice, a Greens Senator and former mayor of Maribyrnong and the couple’s two children. In 2003, Whetton underwent gender affirmation surgery.

Whetton died on 11 September 2019 in Sisters Beach, Tasmania.

In 1989 she was recruited to in the Atmospheric Research division of CSIRO (later changing to CMAR CSIRO Marine and Atmospheric Research). Whetton was promoted to research leader in 1999, and then a research program manager in 2009. Whetton was the Lead Author for the Fourth Assessment Report of the UN Intergovernmental Panel on Climate Change (IPCC) that was presented with the Nobel Peace Prize in 2007.
