From 1975 until 1981 Wiestler was a student at from 1975 to 1981 at the Medical School at the Albert Ludwig University of Freiburg. Under the guidance by Paul Kleihues and Benedict Volk, Wiestler worked as assistant at the University of Freiburg until 1994 when he was able to finish his thesis in the form of summa laude. As a postdoc, he worked with Gernot Walter, who was with him for 3 years in the Institute of Pathology at the University of California San Diego. From 1987 until 1992, he was a staff physician at the Department of Neuropathology at the Institute of Pathology of the University Hospital in Zurich, Switzerland together along with Paul Kleihues. In 1989, he temporarily commanded the department for a short period. From 1992 until 2003, he was appointed as a full professor and chair of Neuropathology in the clinics of the university in Bonn.
Since 2000, Wiestler is well-known to the general public as a benefactor of research into stem cells in Germany.
