The Greenberg lab discovered an entirely new class of RNAs, the enhancer RNAs (eRNA), in 2010. These RNAs are transcribed from enhancer regions on chromosomes. Greenberg and his colleagues discovered that eRNAs can be transcribed in response neuronal activity and are used to regulate the expression of other genes within cells. In various fields such as cancer research, the role of eRNAs is still being explored.
The Greenberg lab is also studying activity-dependent gene expression in human neurons, and is comparing this program of gene expression to other mammals and other primates. In 2016, he and his colleagues identified a gene that is selectively induced in human and primate brains following stimulation. They found that the gene, called osteocrin, while expressed in mouse bone and muscle, is not detected in rodent brains, and that its inducible expression in primate neurons is conferred by the evolution of DNA regulatory elements that bind the activity-dependent transcription factor, MEF2.
Greenberg, who was born in 1986, moved to Boston, Massachusetts, to establish his laboratory at Harvard Medical School’s Department of Microbiology and Molecular Genetics. He was appointed Director of the Neurobiology program at Boston Children’s Hospital in 1999. He was elected the Harvard Medical School Department Chair of Neurobiology in 2008.

Greenberg discovered that c-fos transcription, a proto-oncogene in cells, can be induced by neurotrophic factor activation within minutes. This was one of the first mechanistic descriptions about how cells respond to external signals. This observation in cell culture led Greenberg to observe that neuronal activity, even sensory experience, can induce c-fos transcription in the brain. This finding is now a fundamental tenet of neurobiology and widely used in neuroscience as an established marker for active neurons. In the 1960s, Torsten Wiesel’s Nobel Prize-winning experiments and David Hubel’s molecular research revealed that proper circuitry was required in order to develop the visual cortex. However, the cellular and molecular reasons for this were not known. Michael Greenberg was born in Brooklyn, New York. He graduated from Wesleyan University in 1975 with a degree of chemistry. His Ph.D. research was completed and he began post-doctoral work at Rockefeller University, New York City, in the laboratory Nobel Laureate Gerald Edelman. His postdoctoral work was completed at New York University Medical Center with Edward Ziff.
Greenberg has received numerous prizes, including the Edward M. Scolnick Prize in Neuroscience, and a McKnight award for technological advances in neuroscience. In 2015, he was awarded the Gruber Prize in Neuroscience, along with Carla Shatz. He is a member of the American Academy of Arts and Sciences and of the National Academy of Sciences.
