Sean Ekins was born in Cleethorpes, England, on 2 March 1970, to John Ekins and Elsie May Ekins. He was raised in Grimsby. Ekins later earned the HND Science Applied Biology from Nottingham Trent University (formerly Polytechnic from 1988 to 1991) with a degree in 1991. Ekins later earned his M.Sc. on clinical Pharmacology (1991-1992) from the University of Aberdeen with a dissertation titled “Speculations on the relative roles of cytochrome P450 and flavin containing monooxygenase in the metabolism of S12363” He then received the Ph.D. on clinical pharmacology from the University of Aberdeen in 1996 which was funded by Servier and completed a thesis that read “Maintenance and preservation of xenobiotic metabolism using cryopreservation in precise liver slices. The evaluation of an alternate in vitro method for isolated liver cells”. In the course of his PhD He developed an interest in the prediction of interactions between drugs and drugs using computational methods as an alternative to animal models.
In 2005, he obtained his D.Sc. in science from Aberdeen University. University of Aberdeen with a thesis titled “Computational and in vitro models for predicting drug interactions in humans”.
Since 2006 Ekins has consulted for several companies including on pharmacoeconomics and performing research on Tuberculosis Drug Discovery for Collaborative Drug Discovery.

In the month of December 2001, he began working for a startup company called Concurrent Pharmaceuticals (now Vitae Pharmaceuticals) as Associate Director of computational drug discovery. He was responsible for the development of computational models for ADME/Tox as well as the targets of significance. In this period, he developed an interest in multi-pharmacology of ADME/Tox proteins. In 2004, he joined GeneGo (now part of Thomson Reuters) as Vice President, Computational Biology and developed the MetaDrug product (patent in the process of being filed).

From 1996 to 1998 Ekins continued his studies as a postdoc in the Eli Lilly and Company laboratories studying the little-studied CYP2B6 and applied computational techniques for this enzyme. He gathered drug-drug interaction Ki data for various P450s and created pharmaceuticals. He designed test sets to validate the models, which were later published. He published seminal thoughts about how these methods could be utilized to create collections of compounds to predict interactions between drugs.
In the latter half of 1998, Ekins was hired by Pfizer and continued to pursue his interests of predicting interactions between drugs as well as ADME properties. In 1999, he transferred to Lilly to establish a predictive ADME/Tox research group. Between 1999 and the end of 2001, he developed pharmacophores as well as statistical models for a variety of proteins, including PXR, P-glycoprotein and enzymes.
Ekins has edited or co-edited 4 books for Wiley including: Computer Applications in Pharmaceutical Research and Development (2006), Computational Toxicology: Risk Assessment For Pharmaceutical and Environmental Chemicals(1007), Drug Efficacy, Safety, and Biologics Discovery(2009) and Collaborative Computational Technologies for Biomedical Research (2011) All the books have an underlying connection with computational technologies and their application for pharmaceutical R&D.
