Ron Konopka, a geneticist at the California Institute of Technology, discovered the Period gene and discovered it to be associated with the clock that regulates the circadian rhythm of Drosophila. It was in 1999 that Paul Hardin discovered that per mRNA was subject to intense circadian oscillations after exposure of isolated wild-type permRNA to a sequence of dark-light (LD) cycles that were followed by constant dark (DD). As a postdoctoral researcher in the laboratory of Chronobiologist Dr. Michael Rosbash, Hardin specifically observed the per mRNA levels found in Drosophila brains vary by 10 times in a typical 24 hour light-dark cycle. Hardin also demonstrated that the wild-type PER can restore rhythmic activity in the mRNA of an arrhythmic mutation in the per gene. His research suggest that feedback in the PER protein is responsible for regulating the levels of per MRNA. Hardin later published his seminal study about the specific rhythm of per-mRNA within Drosophila published in Nature. The discovery was the catalyst that led Hardin along with other well-known researchers in the area of chronobiology to create an explanation of the mechanism of clocks in Drosophila. The model is known by”the Transcription Feedback Loop, which implies that the translation protein gives negative feedback to the mRNA transcription process of itself.
Hardin obtained his B.S. in biology from Southern Methodist University (SMU) in 1982. Then he continued to pursue the Ph.D in genetics at Indiana University in 1987 with William H. Klein. He continued to pursue postdoctoral research in Brandeis University under the supervision of the chronobiologist Michael Rosbash. From 1991 until 1995, Hardin was an assistant professor of biology at Texas A&M University, and from 1995 until 2005 , he was a professor at the University of Houston. From 2005 onwards, Hardin has worked as a researcher and professor in the department of biology in Texas A&M University. He teaches classes on basic biology, molecular cell biology, as well as a graduate-level class in biological clocks. Hardin is also director of Texas A&M’s Center for Biological Clocks Research as well as a professor of the Texas A&M Institute for Neuroscience and the PhD programme in genetics. Thus, VRI and PDP1, together with dClock itself, comprise a second feedback loop in the Drosophila clock that gives rhythmic expression of dClock, and probably of other genes, to generate accurate circadian rhythms.
