The family was born at Barambagarh, Odisha, Padmini began playing chess at the young age of nine (2003) due to the father, and Dr. Ashok Kumar Rout’s love to play the game. She attended school at D.A.V. Public School in Chandrasekharpur and she graduated with a degree in commerce in Commerce from BJB College located in Bhubaneswar.

In 2005, Rout was the first woman to win a national title, a title for girls under 11 at Nagpur. She won the title in 2006 when she was in the position of Indian Under-13 Girls champion as well as her Asian girl’s under-12 champion. Rout took home her place in the U14 girl’s section at the Asian in 2006 and the World Youth Chess Championships in 2008. The following year, she placed top of the line with the Asian Junior (Under-20) Girls Championship. The year 2010 was the time she took home in the Indian Junior (U19) girl’s championship and won the bronze medal in both the Asian as well as World Junior Girls Championships.

Rout played for the Indian national team at Women’s Chess Olympiad, Women’s World Team Chess Championship and Women’s Asian Team Chess Championship. She won an individual gold medal playing on the reserve board at the 2014 Women’s Chess Olympiad in Tromsø, Norway. She has been part of the Indian women’s team in the subsequent Chess Olympiads in 2016 at Baku, Azerbaijan and 2018 at Batumi, Georgia.
