Glasspool was ordained on May 15th on the 15th of May, 2010 on the 15th of May in Long Beach, California by the Presiding Bishop of the Episcopal Church, Katharine Jefferts Schori. Glasspool has become the only publicly avowed lesbian chosen to be the first bishop within the Episcopal Church or more general Anglican Communion, and is also the 17th woman who was elected to the episcopate of the Episcopal Church. Her election has garnered international attention as part of the current debate on homosexual bishops within Anglicanism. She was a bishop suffragan for the Episcopal Diocese of Los Angeles from 2016.

She was also the Rector from St. Margaret’s Episcopal Church, Annapolis, from 1992 to 2001, and was also serving as the canon to bishops of Annapolis in the Episcopal Diocese of Maryland from 2001 until 2009.
