She was a member of Ontario’s Legislative Assembly of Ontario from 1995 to 2011 , as an official of the Ontario Liberal Party, serving as Minister during the administration under Dalton McGuinty. She did not run in the 2011 provincial election and took a position as director of business and global markets at PricewaterhouseCoopers. On the 8th of November 2012 Pupatello made public her plans to stand to lead the Liberal Party of Ontario. On January 26, 2013 she was defeated by Kathleen Wynne on the third and final vote. Pupatello was re-elected by a landslide in the 1999 provincial election for the redistributed constituency of Windsor West, while the Progressive Conservatives were re-elected to a second consecutive majority government. Pupatello remained a member of the opposition frontbench, serving over the next four years as Deputy Leader of the Opposition and Official Opposition Critic for Health and Long-Term Care.
The Liberal Party won a majority government in the 2003 provincial election, and Pupatello was again re-elected in Windsor West with a significant majority. On October 23, 2003, she was appointed to cabinet as Minister of Community and Social Services with responsibility for Women’s Issues. Pupatello was elected for the first time as a member of the Ontario legislature during the provincial election of 1995 and defeated New Democratic Party candidate Arlene Rousseau with 5,526 votes in Windsor-Sandwich. The seat was previously in the hands of New Democrat George Dadamo, who was not seeking an election for re-election. It was the Progressive Conservative Party won a majority in the elections in the election, and Pupatello was elected to the legislative chamber as an opposition member of the Provincial Parliament (MPP).
