Fischer was born in Hilversum in 1951. She has an M.A. in the field of History of English and General Linguistics from Newcastle University (1975) and an M.A. (doctoraalexamen) on English Language and Literature from the University of Amsterdam (1976, cum laude). She earned her PhD at The University of Amsterdam (1990, cum laude) with a thesis titled Syntactic Change and Causation: Changes in Infinitival Constructions of English.

Other areas of interest include iconicity and grammaticalization in the language and the relationship between the two phenomena. She is also interested in the ontological significance of grammaticalization, as well as whether it is best to view it as an epiphenomenon, or a distinct mechanism for language change. This issue is discussed thoroughly in her work Morphosyntactic Change (Oxford University Press 2007) in which she examines functional and formal methods of linguistics from the past and changes in language.

Fischer was a professor at the Department of Germanic Linguistics at the University of Amsterdam from 1999 until her retirement in the year 2016. In her academic professional career she was a visiting scholar at from the University of Manchester and a visiting professor at the University of Zurich, the University of Innsbruck, the University of Vienna, and the University of Lille III. Fischer was President of the Societas Linguistica Europaea in 2011. From 2014 until 2016, she was the President of the International Society for the Linguistics of English.
