President Barack Obama nominated Du on August 2, 2011 to replace Judge Roger L. Hunt, who had assumed senior status in 2011. The Senate Judiciary Committee narrowly recommended her nomination to the Senate floor on November 3, 2011. It was voted by 10-8 party lines. Du was nominated by 39 nays to 59 ayes on March 28, 2012. On March 30, 2012, Du was awarded her commission. Du was the first Asian Pacific American to be appointed as an Article III judge for Nevada. On September 2, 2019, she was elected Chief Judge.

In October 2016, Judge Du granted several Nevada Native American tribes a preliminary injunction which required the state to set up early voting and election day polling locations on their reservations. The plaintiffs argued that many of them would have to travel nearly 100 miles round trip to reach polling sites, and that Section 2 of The Voting Rights Act of 1965 required the state to take the location of their reservations into account when planning polling locations. Judge Du agreed in part, granting preliminary early voting and election day relief, but found that the plaintiffs did not have standing to seek in-person voter registration staffing as well.

Du was then a lawyer and represented John Woods in a Truckee Meadows Water Authority lawsuit. Du filed a third-party complaint, despite acknowledging and agreeing to the information in a motion that was contrary to the complaint she had filed. After Valerie Cooke tried to find mishandling of the case, Sandoval signed off as a judge on 2008 sanctions against Du.

Judge Du agreed and ruled that Echavarria was entitled to a new trial.
