Brenton began her political career running for City-County Council of Indianapolis in 1995, running as a Libertarian. She lost, but managed a record percentage of the vote for a Libertarian candidate at the time. It wasn’t until 2016 that she decided to run for office again, this time for United States Senate, against Republican Todd Young and Democrat Evan Bayh. She managed to finish with 5.5 percent of the vote in the general election. She ran for U.S. Senate in Indiana again in 2018, against incumbent Democrat Joe Donnelly and Republican Mike Braun, faring a little worse, finishing with 3.9 percent of the vote.
