Gordon started her comedy career at the age of 16 in New York, where she was employed at the Comix comedy club and hosted live shows alongside Pete Holmes. At the time of her move into Los Angeles in 2010, Gordon was working on comedy and writing freelance full-time.

Gordon also co-wrote the screenplay of and co-executive produced the 2017 film The Big Sick, with her husband Kumail Nanjiani. The critically acclaimed film is based on the beginning of their relationship, with Nanjiani playing himself, and Gordon (renamed Emily Gardner) played by Zoe Kazan. The Big Sick was directed by Michael Showalter and produced by Judd Apatow.

She co-wrote the 2017 romantic comedy film The Big Sick, based on her relationship with her husband and frequent collaborator, comic Kumail Nanjiani. Gordon and Nanjiani won an Independent Spirit Award for Best First Screenplay for The Big Sick; they were also nominated for a 2018 Academy Award for Best Original Screenplay, among many other nominations.

Gordon is the author of two web series targeted at teens, called”Power Up and ExploreD on Disney.com. She was a writer on the second episode of The Carmichael Show on NBC and wrote an episode titled “New Neighbors”. Alongside the writing aspect, Gordon also produces comedy. The year 2010 was the first time she co-created the weekly live show hosted with Jonah Ray and Kumail Nanjiani named The Meltdown with Jonah and Kumail situated in the back of the comic bookshop Meltdown Comics. Gordon was the booker and producer. In 2011, she was contacted to assist Chris Hardwick, who wanted to transform this space into a comedy club, to serve as the show’s program director. She was the program director at Nerdist Showroom from 2011 to 2012.
