Papandrea was born at Adelaide, South Australia, and then moved into New York City in the 1990s, after having a successful profession in the field of filmmaking. In the interim, she returned to Australia for the production of the movie Better Than Sex, which was nominated for an AFI Award for Best Film prior to her move into London after 2001. She worked as an executive in the production department in the movie studio Mirage Enterprises and later returned to New York to work for GreeneStreet Films, a production company that makes independent films, as creative director. At GreeneStreet she was the executive producer of the love story Wedding Daze before joining another independent production company, Groundswell Productions, in Los Angeles in February 2006. She was in charge of the production of Smart People (2008), Milk (2008), The Marc Pease Experience (2009) and All Good Things (2010).

Papandrea made her debut in 2011 with Warm Bodies, a zombie comedy film written and directed by Jonathan Levine and released in 2013. In 2012 she together with actor Reese Witherspoon co-founded Pacific Standard which is a Beverly Hills-based production company focused on the creation of films that are made by and for women. The first two films they made at Pacific Standard were Gone Girl and Wild which were both made from books and rights acquired from Papandrea and Witherspoon before the publication. Both films came out in 2014, at which time they also had completed the production of the comedy Hot Pursuit. In 2016 they announced they would be ending their collaboration however they were going to continue working on projects in the stage of production, such as their HBO Big Little Lies Big Little Lies and a film adaptation of the novel Luckiest Girl Alive.

On 5 February 2019 it was announced that Papandrea will be credited as an executive producer alongside Lesli Linka Glatter and Charlotte Stoudt in the upcoming Netflix thriller series, Pieces of Her.
